The E10S eyepiece with 10x magnification features a graduated scale in the field diaphragm plane. It functions as a standard 10x eyepiece and can also measure the size of objects or their elements. The scale is marked in millimeters and centimeters. To take measurements, you must first calibrate the eyepiece for each objective you wish to use. The E10S eyepiece fits into a 23.2 mm tube.

Calibration

To calibrate the eyepiece, you will need a calibration slide with a scale division of 0.01 mm. Place the slide on the stage and focus the microscope on the scale. Then rotate the eyepiece in the tube until the scale divisions on the slide and the eyepiece are parallel to each other. Finally, perform a calculation.

Calculation

You need to calculate the ratio between the divisions on the eyepiece scale and those on the calibration slide (stage micrometer): how many eyepiece scale divisions fit within the slide scale (for low-magnification objectives), or how many slide divisions fit within the eyepiece scale (for medium- or high-magnification objectives). Then use the formula:
E=TL/A, where:
E is the eyepiece scale division;
T is the stage micrometer scale division marked on the stage micrometer (0.01 mm);
L is the number of marks on the stage micrometer;
A is the number of marks on the eyepiece scale.

Record the number obtained from the calculations for each objective.
The size of the samples is calculated as follows: determine how many divisions the sample occupies on the eyepiece scale. Multiply this number by the result obtained from the formula for each objective. This number represents the sample's linear dimension.
