Description

Calibration slide for microscopes.
The slide has three horizontal micrometer scales with 100 divisions. The division of the scale is 0.01 mm, 0.05 mm and 0.1 mm respectively.
The calibration slide (stage micrometer) is used to determine the magnification, linear field of view of the microscope, and the division value of the eyepiece scales and graticules.
When used in conjunction with a digital camera, the slide allows you to calibrate the image analysis software for each lens.

Calibration of image analysis software

Calibration is performed on each objective used for observations. The calibration slide is used to determine the calibration factor of the image analysis software.

Calibration procedure:

  1. Fix the calibration slide on the microscope stage.
  2. Select the desired lens and set the maximum resolution of the camera.
  3. Get a contrast image of the scale on the monitor screen and capture the image.
  4. Select 'Calibration' mode in the image analysis software.
  5. Click the mouse to define the maximum visible distance between the first and last divisions of the scale.
  6. Enter the resulting scale size value in real-world units into your image analysis software.
  7. Enter the calibration name specifying the current lens magnification.
  8. Check the result: In the 'Measurement' mode, measure in real units a few divisions of the calibration slide scales.
  9. The image analysis software will save the calibration factor under the selected name. It will then automatically convert the results to the selected measurement units.

Calibrating the eyepiece scale

Calibration of the eyepiece scale is performed on each objective used for observations. It is necessary to place the slide on the stage and then adjust the focus of the microscope on the scale. It is necessary to rotate the eyepiece on the optical tube until the scale of the slide and the eyepiece are aligned parallel to each other.

If you use low-power objectives, you need to calculate how many values on the eyepiece scale fit into the slide scale.
If you use medium and high magnification objectives, you need to calculate how many slide values fit into the eyepiece scale.

The CS3 MAGUS calibration slide features three micrometer scales with different division values. Calibration is most convenient when the slide scale covers the entire scale of the eyepiece. With the low magnification objective, a scale with divisions of 0.1 mm or 0.05 mm is used. With objectives with magnifications greater than 20x, a scale with divisions of 0.01 mm is used.

The calculation for each objective is done according to the formula:
E=ТL/A, where
E – eyepiece division value
Т – stage division value specified on the stage micrometer
L – number of divisions of the stage micrometer
A – number of divisions of the eyepiece scale.

The relative values of the eyepiece scale division of each objective will then be recorded.

When measuring an object, you will first need to count the number of eyepiece scale divisions that the object covers. The result will then be multiplied by the scale value previously calculated for the objective in use. The resulting value will be the true linear value of the object.
