The E10S eyepiece with 10x magnification has a graduated scale in the plane of the field diaphragm. It acts as an eyepiece regular with 10x magnification, and is also able to measure the size of objects or their elements. The scale is marked in millimeters and centimeters. To measure, you must first calibrate the eyepiece for each objective you want to use. The E10S eyepiece fits into a 23.2 mm tube.

Calibration

To perform calibration, you will need a calibration slide with a scale division of 0.01 mm. Place the slide on the stage and point the microscope at the scale. Then, rotate the eyepiece in the tube until the slide and eyepiece scale divisions are parallel to each other. Finally, make a calculation.

Calculation

You need to calculate the relationship of the divisions on the eyepiece scale and that of the calibration slide (desktop micrometer): how many divisions of the eyepiece scale fit into the slide scale (for objectives with low magnification) or how many divisions on the slide fit into the eyepiece scale (for objectives with medium or high magnification). Then use the formula:
E=TL/A, where:
E is the division of the eyepiece scale
T is the division of the micrometer scale of the stage marked on the micrometer of the stage (0.01 mm)
L is the number of values on the micrometer of the stage
A is the number of values on the eyepiece scale.

Record the number obtained from the calculations for each goal.
The size of the samples is calculated as follows: it is necessary to determine how many divisions are occupied by the sample on the eyepiece scale. Multiply this number by the result of each goal's formula. This number will reflect the linear value of the sample.
