The SE10S eyepiece not only features 10x magnification, but also a scale to evaluate the size of the sample or its structures in millimeters or microns. The scale is marked with 0.1 mm divisions and is located in the field diaphragm area of the eyepiece. Before you take the measurement, you need to calibrate the scale against the parameters of each lens you want to work with. The SE10S model is installed on a 30 mm pipe.
Calibration
Calibration is performed via a calibration slide (desktop micrometer). Place it on the stage and point the microscope at the scale, divided into 0.1 mm segments. Then, rotate the eyepiece in the tube until the divisions of the eyepiece scale are parallel to the divisions on the calibration scale.
Calculation
To calibrate the eyepiece scale for the selected objective, use the following formula.
E=TL/A, where:
E is the division of the eyepiece scale
T is the division of the micrometer scale of the stage marked on the micrometer of the stage (0.01 mm)
L is the number of values on the micrometer of the stage
A is the number of values on the eyepiece scale.
Write down the E value for each goal. When measuring the sample, count the number of divisions on the eyepiece scale between the two desired points. Then, multiply this number by the value E. The result is the linear value of the measured object.
